I woke up seriously looking forward to my morning CBD oil fix … I mean, tonic. Truth be told, I’m an anxious person. Certain facets of acute cannabinoid exposure, including tachycardia, hypothermia, orthostatic hypotension, dry mouth, ocular injection, intraocular pressure decreases, etc. are subject to rapid tachyphylaxis upon continued administration (Jones et al 1976). No dose tolerance to the therapeutic effects of Sativex has been observed in clinical trials in over 1500 patient-years of administration. Additionally, therapeutic efficacy has been sustained for several years in a wide variety of symptoms; SAFEX studies in MS and peripheral neuropathic pain, confirm that Sativex doses remain stable or even decreased after prolonged usage (Wade et al 2006), with maintenance of therapeutic benefit and even continued improvement.
Because of this inexact fit, the CBD molecule won’t provide the necessary power to turn the neuron on like the THC molecule would. This is similar to the way a AAA battery won’t provide the necessary power that a AA battery would. So once a CBD molecule is seated in the CB1 receptor (not turning it on), the CBD molecule also occupies the space within the neuron and prevents the THC molecule from docking. Because of the way CBD blocks THC from docking in the neuron, it is known as a CB1 antagonist. Just imagine if there was no CBD present. THC molecules would dock with the all the CB1 receptors, turn them on, and take you higher than you’ve ever been before. And before you say, “Right on!”, keep in mind that too much THC—too much psychoactive influence—is called a bad trip. At the worst, it’s called an overdose. Thank goodness CBD keeps this from happening.

I have 4 rescue dogs each with different health problems. I’ve switched completely to homemade food & eliminated numbers outside chemicals & byproducts. The dog suffering from an autoimmune disease is almost completely off of his medication. My other dog who has tumors, the size & adverse effects are deteriorating. I’m thrilled & look forward to improvements with long term use. CBD is such a blessing!!!

Research has shown that administration of cannabidiol actually inhibits agonist effects at the CB1/CB2 receptor sites. Although the effects of CB1 inverse agonism aren’t fully elucidated, many speculate that CB2 inverse agonism may contribute to cannabidiol’s anti-inflammatory effects. Due to the fact that neuroinflammation is associated with anxiety disorders, we could hypothesize that a decrease in inflammation may yield anxiolytic responses in a subset of CBD users.
Long before mechanisms of cannabinoid analgesia were understood, structure activity relationships were investigated and a number of synthetic cannabinoids have been developed and utilized in clinical trials, notably nabilone (Cesamet®, Valeant Pharmaceuticals), and ajulemic acid (CT3, IP-751, Indevus Pharmaceuticals) (Figure 1).
Results indicated that CBD significantly reduced subjective measures of anxiety as evidenced by changes in VAMS scores. Neuroimaging data revealed decreased ECD-tracer uptake when participants received the CBD compared to when they took the placebo. Particularly, activity in the left amygdala-hippocampal complex and the left posterior cingulate gyrus decreased following CBD administration.
Concerns are frequently noted with new drug-drug interactions, but few have resulted in Sativex RCTs despite its adjunctive use with opiates, many other psychoactive analgesic, antidepressant and anticonvulsant drugs (Russo 2006a), possibly due to CBD ability to counteract sedative effects of THC (Nicholson et al 2004). No effects of THC extract, CBD extract or Sativex were observed in a study of effects on the hepatic cytochrome P450 complex (Stott et al 2005b). On additional study, at 314 ng/ml cannabinoid concentration, Sativex and components produced no significant induction on human CYP450 (Stott et al 2007). Thus, Sativex should be safe to use in conjunction with other drugs metabolized via this pathway.
Finally, as we’ve seen in the Cannabidiol Cancer Research article, CBD has been shown to ease pain caused by some chemotherapy drugs (primarily in breast cancer patients), which helps patients tolerate chemo treatments more easily and with less pain-related side effects.
